چکیده

Nanobubbles can be observed with optical microscopy using the total internal re ection uorescence (TIRF) excitation. We report on TIRF visualization using Rhodamine 6G at 5μM concentration which results to strongly contrasted pictures. The preferential absorption and the high spatial resolution allow to detect nanobubbles with diameters of 230 nm and above. We resolve the nucleation dynamics during the water-ethanol-water exchange: Within 4 min after the exchange bubble nucleate and form a stable population. Additionally, we demonstrate that tracer particles near to the nanobubbles are following Brownian motion: A remaining drift ow is weaker than a few micrometers per second at a distance of 400 nm from the nanobubble's center.

Imaging with total internal reflection fluorescence microscopy for the cell biologist.

Total internal reflection fluorescence (TIRF) microscopy can be used in a wide range of cell biological applications, and is particularly well suited to analysis of the localization and dynamics of molecules and events near the plasma membrane. The TIRF excitation field decreases exponentially with distance from the cover slip on which cells are grown. This means that fluorophores close to the ...

Total internal reflection fluorescence (TIRF) microscopy illuminator for improved imaging of cell surface events.

Total internal reflection fluorescence (TIRF) microscopy is a high-contrast imaging technique suitable for observing biological events that occur on or near the cell membrane. The improved contrast is accomplished by restricting the thickness of the excitation field to over an order of a magnitude narrower than the z-resolution of an epi-fluorescence microscope. This technique also increases si...

Visualization of cortex organization and dynamics in microorganisms, using total internal reflection fluorescence microscopy.

TIRF microscopy has emerged as a powerful imaging technology to study spatio-temporal dynamics of fluorescent molecules in vitro and in living cells. The optical phenomenon of total internal reflection occurs when light passes from a medium with high refractive index into a medium with low refractive index at an angle larger than a characteristic critical angle (i.e. closer to being parallel wi...

Reconstituting dynamic microtubule polymerization regulation by TOG domain proteins.

Microtubules (MTs) polymerize from soluble αβ-tubulin and undergo rapid dynamic transitions to depolymerization at their ends. Microtubule-associated regulator proteins modulate polymerization dynamics in vivo by altering microtubule plus end conformations or influencing αβ-tubulin incorporation rates. Biochemical reconstitution of dynamic MT polymerization can be visualized with total internal...

Micro and Nanobubble Water

Nanobubble is a mystery in terms of stability and longevity. Nanobubble, now directly imaged by tapping mode atomic force microscopy or by dynamic light scattering, were previously inferred from long ranged attraction between hydrophobic surfaces immersed in water. They exists in an interconnected, irregular and closed pack structure whose lifetime is in order of hours and whose morphology is d...
